The ArchaeMaxx® Factor Advantage

In addition to higher accuracy, PfuUltra high-fidelity DNA polymerase provides superior overall performance and more robust amplification of longer targets due to Stratagene's patented ArchaeMaxx® polymerase-enhancing factor. The ArchaeMaxx factor improves the yield of products that contain Pfu DNA polymerase by overcoming "PCR poisoning", which is caused by dUTP accumulation during PCR through dCTP deamination2. Incorporation of dUTP inhibits Pfu and many other archaeal proofreading DNA polymerases, limiting their efficiency3. The ArchaeMaxx factor functions as a dUTPase, converting poisonous dUTP to harmless dUMP and inorganic pyrophosphate resulting in greatly enhanced overall PCR performance, including shorter extension times, higher yield and greater target length capability.

Higher Amplification Efficiency

Amplification efficiency is the measurement of fold amplification per PCR cycle, and is expressed as a fraction or percentage relative to perfect doubling (a PCR resulting in perfect doubling would exhibit 100% amplification efficiency). Once the efficiency of a PCR system is determined, it can be used to measure fold amplification from a particular template amount, or alternatively, to calculate the number of PCR cycles needed to obtain a desired yield. A small change in amplification efficiency is magnified over a multiple-cycle PCR, causing significant changes in product yield. For example, in a 25-cycle PCR, a 5% improvement in amplification efficiency can translate to a two-fold increase in product yield.† Lower-efficiency reactions not only produce lower yields, but take longer to reach saturation than high-efficiency reactions, increasing the chances of generating primer-dimers and undesired side products.

Using real-time QPCR methods, it was determined that PfuUltra high-fidelity DNA polymerase exhibits higher amplification efficiencies than other proofreading polymerases4.
 

The Best Enzyme for Cloning

Like Pfu DNA polymerase, PfuUltra high-fidelity DNA polymerase produces blunt-ended PCR products, making it ideal for blunt-ended cloning projects using the PCR-Script® vector or other blunt-ended cloning vectors. This efficient proofreading enzyme simplifies PCR cloning projects by reducing the number of clones that must be sequenced in order to identify an error-free clone and minimizes the need to correct PCR-generated errors by site-directed mutagenesis. For example, using PfuUltra high-fidelity DNA polymerase results in less than a 0.5% chance of error when amplifying a 500-bp fragment, making it the ideal enzyme for PCR cloning. PfuUltra high-fidelity DNA polymerase also amplifies cDNA with high accuracy§.

The accuracy of PfuUltra high-fidelity DNA polymerase surpasses that of all other commercially available DNA polymerases, and is the new benchmark for high-fidelity PCR. The enzyme offers the greatest accuracy possible, while providing robust PCR product yields, high sensitivity, and exceptional target-length capability—making it the superior choice for PCR cloning, site-directed mutagenesis and other high-fidelity PCR applications.

§Amplification of cDNA may require increasing the final Mg2+ concentration from 2 mM to 3 mM.
